Subculture Routine

Resuscitate cells at the higher end of the seeding density (2-9 x 105 cells/ml). When seeding into small volumes, the cell culture flask can be position upright in the incubator. Return to a horizontal position once culture is established.

Upon dilution of cells, seed @ 2-3 x 105 cells/ml and routinely passage twice weekly. Cells can be grown in Corning cell culture flasks and cell factories, however cells can be semi adherent and viability can be reduced when re-suspending cells.

Maintain sub-confluent cultures to retain viability of culture (<9 x 105 cells/ml).

Harvest and passage: remove media carefully (leaving about 0.5-1.0mL) without disturbing cells. Check layer of cells and re-suspend by tapping flask, re-suspend and count.

To prepare antibody: use of cell factories with twice weekly harvest once culture is established.

Cells also grow well in Ultra Low Attachment plastic ware (ULA flasks). Here cells can grow in single suspension and as small cell aggregates.

Growth conditions: 37°C, 5% CO2.

Culture Medium

RPMI + 2mM L-Glutamine + 10% Foetal Bovine Serum (FBS)

Resuscitation: Ensure 20% FBS is used as recover can be very slow. Reduce to 10% once a growing culture is established.

To freeze: 10% DMSO + 90% FBS.
