Cell Line Description

Hybrid generated by fusion of 6-thioguanine-resistant mutant of mouse neuroblastoma N4TG3 (ECACC catalogue number 08062515) and 8-bromodeoxyuridine-resistant mutant of mouse L cell fibroblast line B82 (ECACC catalogue number 08062522) using inactivated Sendai virus. The fusion of N4TG3 x B82 generated two types of hybrid cells: neuronal resembling the neuroblastoma and epithelioid with clearly visible nucleus. 328/7 is of the neuronal type.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:4 to 1:8 i.e. seeding at 2-4x10,000 cells/cm² using 0.05% trypsin/EDTA; 5% CO₂; 37°C.

Culture Medium

DMEM + 2 mM glutamine + 10% Foetal Bovine Serum (FBS) + HAT (0.1 mM hypoxanthine, 10 µM aminopterin and 16 µM thymidine) Cells can be cultured without HAT provided they were cultured in between in HT medium for 3-4 days
