Cell Line Description

Established by SV40 ts30 transformation of human placental cells. The cells express the transformed phenotype at the permissive temperature (33C) and the non-transformed phenotype at the non-permissive temperature (40C). The line has a limited life expectancy of 15 to 18 passages before entering the crisis stage.GMO1

At 40C cells synthesize human chorionic gonadotrophin (hCG) and alkaline phosphatase.

Karyotype

Hypodiploid, modal chromosome number of 42 occuring in 34% of cells. Most cells contained 41-45 chromosomes. Cells with 44 chromosomes also occurred at a high frequency (26%). Cells having a higher ploidly occurred at 20.7%, which is very high.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e. seeding at 3-6x10,000 cells/cm² using 0.05% trypsin or trypsin or trypsin/EDTA; 5% CO₂; 36°C. Change medium 2-3 times weekly.

Culture Medium

Alpha-MEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
