Cell Line Description

Hybrid generated by fusion of bromodeoxyuridine-resistant mutant of rat glioma cell line C6 (C6-BU-1, ECACC catalogue number 08062563) and the 6-thioguanine-resistant mouse L cell fibroblast line A9 (ECACC catalogue number 08062526) using inactivated Sendai virus. Cells formed from such a fusion are multipolar and extend small processes.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm² using 0.05% trypsin/EDTA; 5% CO₂; 37°C.

Culture Medium

DMEM + 2 mM glutamine + 10% Foetal Bovine Serum (FBS) + HAT (0.1 mM hypoxanthine, 10 µM aminopterin and 16 µM thymidine) Cells can be cultured without HAT provided they were cultured in between in HT medium for 3-4 days
