When resuscitating, thaw the ampoule and perform a cell count, spin out the DMSO and resuspend in an appropriate volume of media to give a cell concentration of 4-5 x 105 cells/ml. It may take 2-3 weeks for log phase growth to be achieved. As cells can be slow growing, FBS concentration can be increased to 20% to help establishment.
Doubling time approximately every 4 days. Cells grow singly in suspension. Maintain cultures between 4-9 x 105 cells/ml
Recommended Growth conditions: 5% CO2; 37°C
Culture Medium
RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS). To help establish a culture the concentration of FBS can be increased to 20%.
