Cell Line Description

COS-1 cell line (ECACC catalogue no. 88031701) adapted to grow in the chemically defined protein- and peptide-free InVitrus medium (Cell Culture Technologies). This cell line is substrate dependent i.e. adherent. COS-1 was derived from CV-1 simian cells transformed by origin-defective mutant of SV40 which codes for wildtype T Ag. Possible host for propagation of pure populations of recombinant SV40 virus. The cells should be handled under laboratory containment level 2.

Subculture Routine

In order to prevent syncitia formation, do not allow cells to become confluent. Split semi-confluent cultures 1:3 i.e. seeding at 4x10,000 cells/cm² using an animal component free dissociation reagent; incubate at 37°C. Please note: Once cells have detached from the flask it is necessary to neutralise the dissociation reagent using a compatible inhibitor . Centrifuge the cells; 100-150 x g for 3-5 minutes; count and re-suspend at the recommended seeding density in fresh medium

Culture Medium

InVitrus medium (Cell Culture Technologies http://www.cellculture.com/) + 4mM Glutamine + 5 ug/ml Insulin. When freezing cells down use 50:50 fresh culture medium:conditioned medium plus 10% DMSO.
