Cell Line Description

From pooled male/female gonad tissue of yearling rainbow trout (Oncorhynchus mykiss). First cell line to be established from cold-blooded vertebrates. 8-fold difference in growth rate in temperature range 4-24°C. Susceptible to infectious pancreatic necrosis virus, Egtved virus and arboviruses (EEE, VEE).

Subculture Routine

Passage using 0.05% trypsin below 24°C. Cells may be seeded as few as 3x10,000 - 5x10,000 cells/cm² in 5% CO₂; 24°C. Fish cell lines detach easily during transit if the culture is very young. After resuscitation split cells 1:2 to 1:3 and culture for at least one week with 1-2 media changes before shipping. When cells are growing well they can be split 1:4. Grow cells in the dark (i.e. wrapped in foil if you do not have a suitable incubator). Freeze in FBS+5% DMSO

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).
